Humor, yes, but I would keep it SUPER-kid friendly and toothless. Keep it more on the "I'm fun-loving!" side, and stay away from being cynical and/or ironic.
Another thing might be to make the letter pretty businesslike, but, if you have to include a picture, make it sort of fun - fun, like "wearing a silly hat." I think the idea is to distinguish yourself by pointing out how you're fun and relate to kids, while not crossing the line into someone who would say/wear/do anything inappropriate, or look like someone who doesn't take the job seriously.
